AZD1480 is a novel potent small JAK2 inhibitor with an IC50 of 0.26 nM. Inhibition of Jak2 activity is associated with abrogation of Stat3 nuclear translocation and tumorigenesis. AZD1480 suppresses the growth of human solid tumor xenografts harboring persistent Stat3 activity.AZD1480 inhibited the phosphorylation of Stat5 with an IC50 of 46 nM in TEL-Jak2 cells, whereas little or no inhibition of Stat5 phosphorylation was observed in the TEL-Jak3, TEL-Jak1, or TEL-Tyk2 cells at or below 1 M AZD1480
